The university operates two campuses which extend through two UNESCO World Heritage sites, with its main campus situated in Santiago de Compostela, which serves as the ultimate pilgrimage destination for Camino de Santiago walkers, while its second campus exists within the boundaries of the historic Roman city of Lugo. The university presence consists of its beautiful historical structures and its contemporary research facilities, which operate together under the traditional university motto Sigillum Regiae Universitatis Compostellanae that represents its historical royal and church traditions.

The institution traces its origins back to 1495, when the solicitor Lope Gómez de Marzoa founded a "Grammar Academy" for the poor in the monastery of San Paio de Antealtares. The school established itself as a university during the early 16th century when Archbishop Alonso III de Fonseca supported the institution by creating Fonseca College, which operated as a Renaissance educational facility. The university transformed from a religious education center into a royal educational institution which received the title Regia from King Charles III during the 18th century. The university developed into contemporary scientific disciplines after Jesuits were expelled from their lands, which served as the foundation for today's educational institutions. Throughout the 20th century, USC became a vital catalyst for the cultural revival of Galicia, eventually fostering the creation of other regional universities in A Coruña and Vigo.
