It was founded as the Arellano Law College by Florentino Cayco Sr., a respected Filipino educator and the first Filipino Undersecretary of Public Instruction. The college was named after Cayetano Arellano, the first Filipino Chief Justice of the Supreme Court. The original campus of Arellano Law College was situated in Intramuros, Manila. However, due to the ravages of World War II, particularly the Battle of Manila in 1945, the college was forced to close its doors. It was not until March 1945 that the institution reopened.

In the post-war years, Arellano Law College expanded its offerings and evolved into a comprehensive university. It established new colleges and schools, such as the College of Nursing, the College of Business Administration, and the College of Arts and Sciences. The university's mission was to provide quality education and training to students from all walks of life. Over the decades, Arellano University has continued to grow and adapt to the changing needs of society. It has established additional campuses in various parts of Metro Manila, including Pasay, Pasig, Mandaluyong, and Malabon.
